Study Title Short and long read metagenomics of the 0.2 - 3 micron fraction from the Fram Strait region (2018)
Study Abstract This study incorporates samples collected during the Polarsten PS114 cruise to the Fram Strait region in 2018. Using a CTD Rosette water sample, 1 L of seawater was collected from 10 stations at a depth corresponding to the deep chlorophyll maximum (1 station has two samples from different depths) and sequentially filtered through a 10, 3 and 0.2 micron polycarbonate membrane filter. Samples were divided into two when filtering through the 3 and 0.2 micron filters. DNA was extracted from one of the 0.2 - 3 micron fractions for each station (500 ml seawater). Extracted DNA from all samples was sequenced on an Illumina HiSeq3000 and from three of the samples an additional sequencing was performed on a PacBio Sequel II. The three PacBio Sequel II samples were barcoded and sequenced on a single SMRT cell.
