Description Wetland environments serve as the largest natural source of methane (CH4) to the atmosphere. Hypersaline ponds in the San Francisco South Bay, targets for future wetland restoration, produce methane at rates rivaling those of freshwater wetlands.. This proposal aims to sequence the water, soil, and salt crust microbial communities at a former industrial salt pond in Menlo Park, CA, to better understand the activity and composition of belowground microbial communities, with a focus on methanogenic and methanotrophic microbes, and their anticipated response to wetland restoration. Wetlands in the fresh- and brackish water environments in the SF Bay-Delta region support high rates of aboveground primary productivity, effectively sequestering carbon dioxide from the atmosphere for long-term burial in soils, but also produce methane. The study will help resolve if these methanogen organisms are a) methylotrophic and utilizing non-competitive substrates or b) in the presence of sufficiently low sulfate concentrations can use hydrogenotrophic or acetoclastic pathways for methane production. In addition to sequencing soil cores, this study will incorporate lab-based incubation studies to model the potential response of these microbial communities to future restoration. These incubations will include a mock “freshening” event by incubating soil cores with artificial seawater in a dilution series (240, 120, 60, 35 ppt). The sequencing of these soil microbial communities along a mock-restoration time series will provide an unparalleled opportunity to simulate soil microbial response to future restoration work in the South Bay.
