Description The overwhelming majority of carbon and nutrient input in freshwaters is hypothesized to come from terrestrial sources. However, the source of utilizable carbon is thought to shift from terrestrially derived to phytoplankton derived as the environment becomes more saline. This project seeks to characterize the organic matter cycling mechanisms of active bacteria and their associated viruses along a freshwater to saline gradient in the Delaware River/Bay transition zone. Dissolved organic matter undergoes drastic changes along estuarine mixing, probably resulting in differences in cycling mechanisms and microbial function. We see a gradual shift of the microbial community associated with the salinity gradient along the Delaware Bay, which we hypothesize is greatly influenced by phytoplankton-derived C in the lower part of the bay and by terrestrial C in all sections of the bay. Specifically, we expect to find when genes for carbon utilization are utilized, how they are linked to the type of dissolved organic matter and obtain data on the abundance and use of pathways involved in C1 metabolism (methane, methanol and CO) This research would greatly enhance our knowledge on the diversity, abundance and utilization pathways for organic carbon as they vary over the seasons.
