Orange, white, and yellow vacuolated Beggiatoaceae filaments are visually dominant members of microbial mats found near seafloor hydrothermal vents and cold seeps, with orange filaments typically concentrated toward the mat centers. No marine Beggiatoaceae are yet in pure culture, but evidence to date suggests they are nitrate-reducing, sulfide-oxidizing bacteria. For genomic sequencing, a single orange filament was hand-picked from a Guaymas Basin microbial mat sample and epibionts removed by passage through soft agar. Filament DNA was amplified with phi29 polymerase and the amplified DNA checked for genetic purity by amplification and sequencing of the 16S-23S rRNA spacer region and the 16S rRNA gene. A 454 paired-end library was prepared and sequenced on a GSFLX titanium sequencer (454 Life Sciences). Assembly was performed using the Celera assembler and annotation was done via the Newbler pipeline. A total of 99.3% of the sequence was assembled into 822 contigs, suggesting good coverage was achieved. A total of 4.7 Mb of sequence was recovered with 80% of the sequence forming large (=15 kb) contigs.
